What is the TRiP Screening Center?
The TRiP facility at Harvard Medical School maintains the
entire collection of TRiP stocks for people wishing to
screen the lines en masse.
The facility is located on the third floor of the New
Research Building (NRB) on the Harvard Medical School
campus, in space contiguous with the DRSC. In October 2008,
the TRiP screening facility collection included over 1,200
TRiP stocks and is growing by approximately 250 stocks per
month.
The TRiP screening facility provides the space and
equipment to carry out a complete survey of the current set
of TRiP stocks. If you are interested to screen, please

What TRiP stocks are available in the Screening Center?
All stocks generated in the TRiP are available at the
Screening Center. This includes all of the lines
available at BDSC (lines generated in the VALIUM10 vector)
and approximately 600 stocks lines generated in the
VALIUM1 vector. For maximal knock-down using the TRiP
lines made in VALIUM1, it is best that your GAL4 also
contain UAS-dicer2.
The TRiP team is currently generating about 250
transgenes per month. As they are ready, the
corresponding fly lines will be sent to the BDSC for
distribution.
What do I need to do to work at the TRiP Screening Center?
Contact TRiP Screening Center to reserve a
screening space. At this time you will need to
discuss specifics of the your screen so that the
TRiP Team will be able to meet your screening needs.
Items to consider include any special reagents
and/or equipment needed, method of phenotypic
readout, etc.
Three to four months before arriving at the TRiP
Screening Center:
If screening TRiP stocks in the VALIUM1 vector,
prepare your GAL4 line in a UAS-dcr2 background.
At present approximately 600 TRiP stocks are in the
weaker of the TRiP vectors (VALIUM1). To achieve
maximum knock down your GAL4 line should also carry
UAS-dcr2. It may take up to 2 months to construct your
GAL4; UAS-dcr2 stock so you should plan for this event
ahead of the time your TRiP screen will start.
Two months before arriving at the TRiP Screening
Center:
Send all fly stocks the screener will require for
screening (e.g., GAL4 lines) to the TRiP Screening
Center. Once these stocks arrive at HMS members
of the TRiP team will put the lines through
quarantine (1 month required) as well as amplify
them (1 month required). This will ensure that the
screener will be able to initiate their screen
immediately upon their arrival at the screening
center.
Please Note: All stocks brought into the TRiP
Facility and Screen Center are required to go
through quarantine! No exceptions!
How much will it cost to screen the TRiP stocks?
While the cost for screening this collection at HMS has
not been determined, a close estimate will be $2.50 (US
dollars) per line. By far the greatest expense will be
traveling to and living in Boston.
Note: If needed, members of the TRiP will be
available to answer questions and provide suggestions for
possible housing options in Boston. However, it is the
responsibility of the screener to arrange for housing.
